The active ingredients of Scutellaria baicalensis Georgi, a valuable traditional Chinese medicine, are polyhydroxyflavones, namely baicalin, scutellarin and wogonoside. However, information about the metabolic routes, metabolites and even more the effect of chemical structure on the stability of the three has been limited. In this article, the three natural compounds were incubated with human fecal flora, respectively, and highly sensitive and specific ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry implementing the MetaboLynx? software method was used for the drug metabolism study. The chromatographic separation was performed on a 1.7-μm particle size Syncronis C18 column using a gradient elution system. The components in the extract were identified and confirmed according to the mass spectrometric fragmentation mechanisms, MS/MS fragment ions and relevant literature by means of electrospray ionization mass spectrometry in negative ion mode. With this method, a total of ten metabolites were identified based on MS and MS/MS data. The results indicated that hydrogenation, methylation and deglycosylation were the major metabolic pathways of the three flavone glycosides in vitro, and the metabolic stability was closely related to the chemical structure. This study will be helpful for fully understanding the impact of intestinal bacteria on these active components. Furthermore, this work demonstrated the potential of the ultra-performance liquid chromatography/quadrupole time-of-flight mass spectrometry approach with MetaboLynx for quite rapid, simple, reliable and automated identification of metabolites of natural products.  相似文献

In this paper, we consider a cholera model with vaccination. The disease‐free equilibrium of the system is globally asymptotically stable when the basic reproduction number . If , the disease persists and the unique endemic equilibrium is globally asymptotically stable in the interior of the feasible region under some conditions, which is obtained by compound matrices and geometric approaches. We perform sensitivity analysis of on the parameters in order to determine their relative importance to disease transmission and prevalence. 对强飞秒激光聚焦在空气中所激发的等离子体的发射光谱进行了实验研究.结果表明,光谱特征表现为短波段(截至波长为340 nm)强烈的连续谱和长波段(波长在800 nm附近)强度相对较低的线光谱.在脉冲宽度(50 fs)保持不变而不断调节激光脉冲能量时,等离子体光谱形状的特征基本相似;当激光脉冲能量(1 mJ)保持不变而脉冲宽度从50 fs增加至500 fs和1 ps时,连续谱的峰值(500 nm)显得格外突出,并开始呈现出线光谱特征. 关键词: 飞秒激光 激光空气等离子体 发射光谱 线光谱  相似文献

The monocot lectin from the tubers of Arisaema erubescens (Wall.) Schott has been purified by consecutive hydrophobic chromatography and ion exchange chromatography methods. The molecular weight of this A. erubescens lectin (AEL) was determined to be about 12 kDa by high performance liquid chromatography (HPLC) and sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) methods. AEL could agglutinate rabbit erythrocytes. The haemagglutination activity of AEL was only inhibited by asialofetuin, while monosaccharide did not react. Rat paw edema and neutrophil migration models were used to investigate the pro-inflammatory activity of AEL. AEL (100 and 200 μg/paw) could induce significant rat paw edema. In addition, AEL (100, 200 and 300 μg/mL/cavity) could induce significant and dose-dependent neutrophil migration in the rat peritoneal cavities. Besides, AEL at doses ranging from 100 to 300 μg/mL/cavity could significantly increase the concentration of nitric oxide (NO), prostaglandin E(2 )(PGE(2)) and tumor necrosis factor alpha (TNF-α) in peritoneal fluid. As compared with control animals, 75% depletion in the number of resident cells following peritoneal lavage did not reduce the AEL-induced neutrophil migration. However, pre-treatment with 3% thioglycollate which increased the peritoneal macrophage population by 201%, enhanced the neutrophil migration induced by AEL (200 μg/mL/cavity) (p < 0.05). Reduction of peritoneal mast cell population by chronic treatment of rat peritoneal cavities with compound 48/80 (N-methyl-p-methoxyphenethylamine with formaldehyde) did not modify AEL-induced neutrophil migration. The results provided the basis for identifying the toxic components of A. erubescens and AEL could be a new useful tool for pro-inflammatory research.  相似文献

利用水热合成和无机溶胶法,分别制备了具有棒状(TiO2-R)和无规则结构(TiO2-I)的锐钛矿相TiO2,并以之为载体制备得到Pd/TiO2电催化剂.循环伏安测试显示,与无规则TiO2相比,具有棒状结构的TiO2载Pd催化剂对甲酸氧化的电催化性能提高了70%;计时电流测试显示,运行3000 s后,甲酸在棒状TiO2载Pd催化剂上的氧化电流是无规则TiO2载Pd催化剂的16倍.其原因可能与TiO2纳米棒拥有更好的电子传导性且表面拥有较多的活性含氧基团有关,从而能够有效提高催化剂对甲酸氧化的电催化活性和抗毒化性能.  相似文献

利用法国梧桐絮(花粉)的管状结构,通过简单的碳化处理制备了碳微米管。并用扫描电子显微镜、X-射线衍射光谱仪、拉曼光谱仪和X射线光电子能谱仪对碳微米管进行了详细分析。碳微米管的外径为20~30μm,壁厚约为1μm。其内外壁存在着许多微孔和介孔,BET比表面积为265 m2.g-1。碳微米管具有典型的电化学双电层电容性能,表现出优异的稳定性,展示了在超级电容器方面的应用潜力。  相似文献
